I don't know how to block a number, but I have found a neat way to ignore it.
Somehow, someone in Jamaica, got my number and has been calling me constantly to scam me. After telling him that he was wasting my time and his, He didn't care and continued to call to bother me and just laugh.
So I recorded a ring tone using voice memo - a soundbite with the phone between two pillows. I named it 'Silent' and uploaded it through iTunes to my ringtones
I could tell that he tried to call for several more days by seeing it in my missed phone call section but his calls did not bother me and so he was finally discouraged.
Hopefully this idea will help solve your problem.

You might find this tutorial on how to block calls on iPhone helpful or frustrating. Either way, I think it should provide you with a comprehensive answer to your question.

...........From my saved info I have.
The iPhone doesn't support call blocking, the App Store has no useful apps and you're not looking to jailbreak your iPhone. Where do you go now? Using GarageBand to create a free iPhone ringtone, I've created a silent ringtone, which you can download for free from everythingiCafe. Download and drag this to the Tones section within iTunes and sync to your device.
On your device, go to Phone > Recent Calls > Select the arrow at the right. Edit them as a new contact, scroll down and set their ringtone to the newly synced 'iPhone Silent Ringtone'. Press 'Done'. When they call again, you won't be interrupted. Your display will light up, but that's the worst of it.
Before I ran across this post, I purchased a silent ringtone for 99 cents. Best $ I've spent. When someone I don't want to call again calls, I add that number to my 'contact list' which is named Blocked. I had a rash of robocalls for various schools and they all were added to my Blocked ringtone. Did the trick.
Hope this helps.

You may be able to get a number blocked through your carrier. Sprint offers this to all customers. You can set up a blacklist of numbers at the Sprint site for your account. No text messages, no mms messages, no phone calls. BAM!

Sorry for the length of my silence, but I posted that before work this morning and when I got home from work there was an iPhone 5 (happy face) waiting for me so I've been messing with setting that up.
No, I'm not jailbroken but I don't need to block any calls currently, so my solution works great for me.
